Interference of Corynebacterium pseudotuberculosis with caprine tuberculosis diagnostics tests

C. pseudotuberculosis is the causative agent for caseous lymphadenitis in livestock and some flocks are seriously affected. It has antigenic similarity with species included in Mycobacterium tuberculosis complex. Little is known about the interference between animals infected with this disease or vaccinated against it in diagnostic tests for caprine tuberculosis. Thirteen goats were selected from a tuberculosis-free flock not vaccinated. Seven were sensitized with inactivated bacteria (3 ml of a suspension of 109 CFU/ml). Each animal received two injections (intramuscular and subcutaneous) separately fifteen days. Six non-inoculated animals were kept as controls. Before the sensitization animals were subjected to single intradermal tuberculin (SIT) test, single intradermal cervical comparative tuberculin (SICCT) test and interferon-gamma (IFN-y) assay. Both groups were negative to all tests (for intradermal testing severe interpretation was used during the study). From the day of sensitization until 12 weeks later blood samples were taken, IFN-y showed negative results throughout the study. Five weeks after the inoculation intradermal tests were performed. Four animals out of seven sensitized were positive (SIT test), six out of seven sensitized were positive (SICCT test). Nine weeks later both tests were repeated, one animal out of seven sensitized was positive (SIT test), two out of seven sensitized were positive (SICCT test). The sensitization caused diagnosis interference with intradermal tests. The higher number of reactors when they were performed five weeks post inoculation could be explained to a booster effect in the immune response due to the inoculation. Diagnosis interference in the IFN-y was not detected
