Expression of the a1 integrin subunit in canine gastric carcinoma

Integrins are a superfamily of glycoprotein involved in the binding of cells to the extracellular matrix. They are heterodimers containing two distinct chain types, the ? (alpha) subunit and the ? (beta) subunit. The integrin subunits are in the plasma membrane and have very short cytoplasmic domains. The ?1 integrins possess a domain bound to collagen and the membrane protein laminin (?1 ?1). In addition, aberrant expression of integrins, a group of extracellular matrix receptors has been shown to be involved in the different phases of tumor metastasis. Gastric carcinoma accounts for 60-70% of gastric neoplasms in dogs. In our study we included stomach samples from 25 dogs from the VetPatologos laboratory and processed at the Pathology and Forensic Veterinary Service of the VISAVET center. Histopathologically, they were classified according to WHO criteria, analyzing the malignancy criteria. The most common subtype was signet ring cell carcinoma (SCC) (52%, n=13), followed by undifferentiated (28%, n=7) and tubular (24%, n=6). In the immunohistochemical study, ?1 integrin subunit expression was detected in tumor cells and categorized using the gradation: 0 (absence), 1 (mild), 2 (moderate) and 3 (intense expression). It was found that in SCC and tubular carcinomas, tumor cells had lower immunoreactivity (1-2) than normal glands (2), whereas undifferentiated ones showed higher immunostaining (3). Expression was also higher in tumor cells infiltrating deep and metastatic layers than in more superficial cells. These data may provide interesting information about the involvement of the ?1 subunit in the biological behavior of each type of gastric carcinoma, as well as in the phenomenon of metastasis
