Work Package 1. Management and coordination.

Objectives:
- To provide resources for effective coordination of all project activities and maintenance of contact with Commission representatives.
- To implement an administration and communication infrastructure to establish a basis for efficient and easy communication within the project.
- To effectively monitor the project, in administrative and financial terms.
- To guarantee the adherence of the work to the overall project plans, available resources and planning.
- To undertake quality assessment of project progress, results and impact.
- To ensure that the required reporting is prepared and delivered in a timely manner.
- To ensure that the Description of Work and the Consortium Agreement are maintained and updated where and when necessary.
- Overall legal, contractual, ethical, financial and administrative management of the project.

Work Package 2. Vaccine optimization and scale-up.
WP leader: R. Juste (NEIKER)
Objectives:
- To produce a bank of strains seeds from a selected isolate of M. bovis
- To standardize the medium-large scale production of the selected vaccine strain in terms of shortest time, maximum amount and optimal and stable genetic and antigenic features.
- To demonstrate that DIVA reagents developed in VLA to distinguish BCG vaccination from M. bovis infection can also be applied to this prime-boost scenario
- To standardize the composition of the vaccine product according to the route of administration in terms of dose and adjuvants for maximal protection and minimal diagnostic interference.
- Analysis of commercial aspects for technology transfer.

Work Package 3. Evaluation of vaccine-associated diagnostic strategies including use of defined diagnostic antigens.

Objectives:
- To validate vaccine-associated diagnostic strategies that will allow the differentiation of ‘accidentally’ vaccinated domestic animals from their infected counterparts.
Tasks:
- Task 3.1 Tuberculin skin test and IGRA reactivity
- Task 3.2 Antigenically defined skin test and IGRA reactivity
- Task 3.3 Serological tests in pigs
- Task 3.4 Multi-species serological assay
Work Package 4. Integrated field TB control in wild boar.
WP leader: C. Gortázar (UCLM).
Objective:
- To assess efficacy of oral vaccination alone.
- To assess efficacy of selective culling alone.
- To assess efficacy of the two combined strategies, selective culling + vaccination.
Tasks:
- Task 4.1 Controlled field assessment of oral vaccination in wild boar
- Task 4.2 Test and cull strategies in wild boar captured under field conditions
- Task 4.3 Assessment of the combined effect of previous tasks
Work Package 5. Evaluation of efficacy of heat-inactivated M. bovis as oral vaccine in domestic and feral pigs.
WP leader: P. Pasquali (ISS)
Objectives:
- To assess the efficacy of such vaccine to protect domestic and feral pigs in such endemic area of tuberculosis.
Tasks:
- Task 5.1 Assessment of immune status of vaccinated piglets.
- Task 5.2 M. bovis and corresponding lesions assessment.
Work Package 6. Dissemination, exploitation, market preparation and scientific coordination.
