Proteomic characterisation of bovine and avian purified protein derivatives

Tuberculosis in cattle is subjected to compulsory eradication programmes based on the test-and-slaughter policy. The official diagnostic tests, single intradermal tuberculin (SIT) test and interferon-gamma (IFN-γ) assay, depend on purified protein derivative from Mycobacterium bovis (bPPD). Because of proteins shared with non-tuberculous mycobacteria, the assay specificity requires that the reactivity against bPPD is compared with reactivity against PPD derived from M. avium (aPPD). Recently, to detect positive animals missed by the official tests some research has been centered on the humoral response to mycobacteria. Improving diagnostic tests based on bPPD and aPPD requires detailed understanding of their protein composition, which would allow these reagents to be standardised and further optimised to reduce cross-reactivity. In this study we have done the proteomic characterization of bPPD and aPPD obtained from CZ Veterinaria which are widely used in European bovine tuberculosis eradication programmes. We also performed proteomic analysis of P22, a novel protein complex affinity-purified from the bPPD preparation, which may serve as an alternative antigen in tuberculosis immunodiagnosis.
