Use of antigenic cocktails in the skin test for in vivo diagnosis of bovine tuberculosis

Standard tests used worldwide for diagnosis of bovine tuberculosis are intradermal tuberculin tests and the interferon-gamma assay. The main reagents currently in use are purified protein derivatives (PPDs) which are obtained from Mycobacterium bovis and consist of a poorly characterized protein mix. The PPDs are difficult to standardize, so to overcome this issue some synthetic molecules specific to the M. tuberculosis complex are being studied. The objective of these antigens is also to improve sensitivity and specificity. We have assessed the biological potency of two antigenic peptides and proteins cocktails containing ESAT-6, CFP-10 and Rv-3615c in comparison with bovine PPD. The study was performed in cattle from a herd with natural tuberculosis infection (confirmed by the presence of lesions compatible with tuberculosis and/or microbiological culture). Animals (n=23) were simultaneously inoculated 0.1 mL by injecting PPD and the peptide and protein cocktails using Dermojet system at different sites of the upper neck. For the single intradermal tuberculin (SIT) test the severe interpretation was used following Council Directive 64/432/EEC. Number of reactors obtained was 60.87% (SIT test with bovine PPD), 47.83% (SIT test with peptide cocktail) and 60.87% (SIT test with protein cocktail). Thirteen out of 14 same reactors were identified with bovine PPD and the protein cocktail, and mean increase in skin thickness, 4.6 and 4.2 mm, respectively, was obtained. Results show a potential use of these synthetic reagents for diagnosis, similar to that achieved with standard PPDs although more field studies should be carried out to confirm these results
